Screening the effect of some veterinary antibiotics on the activity of glutathione-s-transferase

In this study, the inhibitory effects of Norocilin, Ceftiofur, Marbocyl,tylan and oxytetracycline antibiotics on GST enzyme activity was reported. It has been investigated for the first time how these antibiotics affect the activity of the GST enzyme in vitro. All antibiotics showed a significant decrease in enzyme activation.

Antibacterial activity of camel milk and whey in combination with some antibiotics against vancomycin resistant Staphylococcus aureus

The main goal of this research is to observe the antibacterial activity of camel milk and whey in combination with clindamycin, doxycycline, meropenem, trimethoprim sulfamethoxazole and vancomycin antibiotics against Vancomycin resistant Staphylococcus aureus (VRSA) and Staphylococcus aureus (ATCC 6538P). Micro-broth dilution method was used to measure the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) for all antibiotics, milk and whey samples. For further antibacterial investigation, the fractional inhibitory concentration (FIC) was also determined. The interpretations of the activity of camel milk and whey combined with the five examined antibiotics produced remarkable synergistic activity in most combinations against both VRSA and S. aureus. It is noticeable that camel milk showed a synergistic effect with clindamycin, doxycycline and trimethoprim sulfamethoxazole against VRSA and S. aureus. Moreover, camel whey exhibited a synergistic effect with clindamycin and doxycycline against the two isolates. The present research clearly showed that camel milk is considered a promising natural product that plays an important role in elevating the susceptibility of bacteria to some antibiotics.

Inhibitory Effect of Meropenem Antibiotic on the Activity of Glutathione-S-Transferases

The following experiment investigated the effect of meropenem antibiotic on the activity of glutathione-S-transferase (GST) isoenzymes. This effect was estimated spectrophotometrically at different concentrations of meropenem using 1-chloro-2,4-dinitrobenzene as a substrate. The obtained results revealed that meropenem antibiotic demonstrated an inhibitory effect on GST activity at low tested concentrations (0.25, 0.5, 1 and 2.5 mM) that resulted in 30.5, 39.4, 45.1, and 100% inhibition, respectively. Also, results provided that the meropenem concentration required to reduce the GST activity to half equal to 1.266 mM. After applying the Line weaver-Burk blot and equation, it was noted that both Km and Vmax values decreased in the presence of meropenem, indicating uncompetitive inhibition. In conclusion, meropenem antibiotic is a potential inhibitor for GST. Therefore, it will be crucial to consider the metabolic defense systems when selecting the dosages of this antibiotic to be utilized for the treatment of infections.

Antibacterial activity of peptides from Rosmarinus officinalis leaves in combination with sulfamethoxazole and trimethoprim antibiotics

Plants are a major source of antimicrobial peptides. So, the aim of this study was to determine the antibacterial activity hof peptides isolated from Rosemary (Rosmarinus officinalis) leaves. Such as activity was compared with the synergistic effect of these peptides in combination with sulfamethoxazole and trimethoprim antibiotics against Escherichia coli, Klebsiella pneumoniae, Proteus vulgaris, Pseudomonas aeruginosa, and Staphylococcus aureus. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) for each combination were measured using the micro-broth dilution method. For further antibacterial investigation, the fractional inhibitory concentration (FIC) was also used. The obtained results demonstrated the antibacterial potential of rosemary peptides against all tested bacteria. Moreover, results showed a strong inhibitory effect against Gram positive bacterium S. aureus with MIC value of 1.563 mg/mL. Also, MBC results provided that S. aureus was the most sensitive one to rosemary peptides with MBC value of 12.5 mg/mL. The interpretations of the activity of rosemary extract combined with both sulfamethoxzole and trimethoprim produced a remarkable synergistic activity against S, aureus, E. coli and P. aeruginosa. In conclusion, the present research clearly showed that rosemary peptides considered as promising antibiotic compounds that play an important role in the enhancing of susceptibility of some antibiotics

Evaluation of Total Phenol Content, Antibacterial and Antioxidant activity of Leaf Aqueous Extracts of Salvia viridis and Senna italica from Palestine

The purpose of this study was to determine the total phenolic content, antioxidant and antibacterial activities in the aqueous extracts of Salvia viridis and Senna italica. The first part of the current research was to estimate the total phenolic content in the tested extracts by using the Folin-Ciocalteu assay. The obtained results indicated that the amount of total phenol in S. viridis and S. italica aqueous extracts is respectively equal to 47.75mg/g and 61.58mg/g in GA equivalent. The other aim of the present project was to determine the antioxidant activity of the inspected plant species through DPPH assay. Results provided that the aqueous extract of S. italica had high antioxidant activity which was similar to the antioxidant activity of ascorbic acid (72.23%) at 50μg/ml. While, the antioxidant activity for S. viridis at 50μg/ml was lower than that of ascorbic acid (42.4%). The last part of this study was to evaluate the antimicrobial efficacy of S. viridis and S. italica against Escherichia coli, klebsiela pneumoniae, Staphylococcus aureus, Bacillus subtillus. The MIC results indicated that S. viridis exhibited the best inhibitory behavior than S. italica. As it inhibited the growth of all tested bacterial isolates at 1.563mg/ml except B. subtilis. The obtained MBC results confirmed that the plant extract from S. italica was the most potent one as it had a bactericidal effect at 12.5mg/ml against E. coli and S. areus. In conclusion, this study supports the traditional medicinal uses of the wild plants in disease therapy.

Screening of anticancer potential of leaf aqueous extract from

Pulicaria incisa growing wild in Palestine

The anticancer potential of Pulicaria incisa aqueous extract was estimated by measuring its cytotoxic and cytostatic effect on human cervical cancer (Hela), human colorectal adenocarcinoma (HT-29), and skin melanoma cancer (B16–F1) cell lines. This effect was investigated by the exposure of cancer cell lines to the aqueous extract of P. incisa at different concentrations (5, 2.5, 1.25, 0.625, 0.3215 mg/mL) for 24 and 72 hours. The results showed that the studied extract inhibited the growth of the various cell lines, eventually leading to cell death, as shown by both MTT and LDH assays. The extract of P. incisa displayed the strongest cytotoxic effect against HT-29 cells at high concentrations at which viability was nearly zero %, while the strongest cytostatic effect of the prepared extract was on the B16–F1 cells at high concentrations. Nonetheless, P. incisa extract gave exciting results on Hela cells with a cytotoxic effect at high concentrations and a cytostatic effect at low concentrations. In conclusion, the cytotoxic and cytostatic effect of P. incisa aqueous extract against the three cell lines is due to the presence of several phytochemicals. In Vitro Activity of Some Medicinal Plants on Blood Coagulation

Objectives: The concern for finding natural and curative agents without adverse side effects has prompted the interest in discovering hemostatic agents from plants. Therefore, in vitro activity of Aizoon hispanicum L. (Aizoaceae), Centaurea hyalolepis Boiss. (Asteraceae), Heliotropium maris-mortui Zohary. (Boraginaceae), Parietaria judaica L. (Urticaceae), Polygonum arenarium Waldst. & Kit. (Polygonaceae), and Verbascum sinuatum L. (Scrophulariaceae) on blood coagulation was estimated by two common tests, which are the prothrombin time test (PT) and the activated partial thromboplastin time test (aPTT).

Materials and Methods: The extracted powders from the plants under this study were adjusted to be 50 mg/mL. Then, in vitro effect of these extracts on the platelet poor plasma samples was measured by an automated coagulation analyzer using PT and aPTT tests.

Results: Based on the obtained results, all plant extracts affected the coagulation cascade by rising either PT or aPTT or both, except for V. sinuatum extract, which reduced only aPTT value. Moreover, the recorded PT values showed that A. hispanicum, H. maris-mortui, and P. arenarium significantly prolonged the PT (p<0.05). Additionally, the results clearly showed that V. sinuatum acted as a coagulant agent based on aPTT values, while all other plants, in contrast, acted as strong anticoagulants. Among the plant species under study, A. hispanicum, H. maris-mortui, and P. arenarium extracts prolonged both PT and aPTT significantly (p<0.05). This could be referred to their additional effect on the common pathway. However, C. hyalolepis, P. judaica, and V. sinuatum showed no significant effect on PT values (p>0.05).

Conclusion: The positive recorded data from this research could serve as identification of new hemostatic remedies that could be used for the commercial economic purposes and for managing several cardiovascular diseases.

Bioactivity of three medicinal plants leaf aqueous

extracts growing wild in Palestine

The aim of the present study was to investigate some biological activities of Aizoon hispanicum, Heliotropim maris-mourtui and Polygonum arenarium which are wildly growing in Palestine. The first part of the current research was to find out the effect of the screened plant extracts on hepatic GSTs spectrophotometrically. The obtained results revealed that all examined different aqueous extracts from the plant species under study demonstrated inhibitory effect on GSTs activity at all tested concentrations. Among all the investigated plant extracts, result showed that A. hispanicum exhibited the strongest inhibitory effect on GSTs activity. The second part of this study was to confirm the antioxidant activity of the investigated plant species through DPPH assay. P. arenarium aqueous extract showed the highest antioxidant activity which is relatively similar to the antioxidant power of ascorbic acid. The last part of this study was conducted to evaluate the antibacterial effect of the three extracts against six different bacterial isolates using broth dilution method. The obtained antibacterial results indicated that all examined extracts were able to act against all studied bacteria with observed variation among them. In conclusion, results indicated that the extracts of the tested plant species could be explored as natural remedies for health promotion

In vitro Screening of Glutathione- S -Transferase Inhibition by Different Extracts of Verbascum sinuatum

Background and Aim: The concern toward using herbs as enzyme inhibitors such as glutathione-s-transferases (GSTs) may result in the improvement of proliferative diseases. Moreover, it may overcome the problem of multidrug resistance tumors. Based on that, this experiment was conducted to detect the impact of using aqueous, ethanol and methanol extracts from Verbascum sinuatum L. (Scrophulariaceae) that grow wild in Palestine as enzymatic inhibitors.

Materials and Methods: The impacts of these three extracts at four studied concentrations (250, 500, 750 and 1000 μg/mL) on the activity of purified hepatic glutathione-s-transferases were estimated spectrophotometrically by using-chloro-2,4-dinitro-benzene (CDNB) as substrate.

Results: The results of the present research indicated that all the three prepared extracts of V. sinuatum could inhibit the activity of GSTs at all examined concentrations. Moreover, according to the results, the alcoholic extracts from the studied plant species were more influential in the activity of GSTs than the aqueous ones . However, minor inhibitory variations were observed between methanol and ethanol extracts. It is notable that all the studied extracts manifested inhibitory effects in a dose-dependent manner.

Conclusion : The results of this study emphasized the possibility of utilizing V. sinuatum extracts in pharmaceutical industry of new medications to fight drug resistant tumors in general and GST-induced tumors in particular.

In vitro Anticancer Effect of Different Extract Types of Ephedra aphylla and Ephedra foeminea on HeLa Cells

Statistics indicate that millions of people are diagnosed with cancer annually. There are many curing cancer protocols, never the less, have low success rate and are still associated with high mortality. Cancer deaths were 14.7% of the total reported deaths in Palestine recently. Asthe utilization of complementary and alternative medicine in Palestine is very common, accordingly, the discovery of novel effective compounds could be achieved by exploring unknown folk medicinal wild plants. The clinical interest in the folk use of Ephedra increased during the 20th century especially for the treatment of cancer.From this point, the ongoing research project issue has emerged, which is to investigate the antitumor effect of aqueous, methanol and ethanol extracts of both Ephedra aphylla and Ephedra foeminae(Gymnosperms) on HeLa cell line in Palestine. As no previous studies on that issue were conducted. Aqueous, methanol and ethanol extracts were prepared at different final concentrations 4,2, 1, 0.5 and 0.25 mg/ml. Both cytotoxic and cytostatic effects of all extract’s types were investigated using inverted microscopy morphological screening and MTT assay under final treatment concentrations equal to 400, 200, 100, 50, 25 µg/ml. The acquired results revealed morphological alteration in which cancerous cells became shrunken, rounder and detached in comparison to the normal and negative controls and MTT cell viability decreasedat all examined plant species different extract types all screened concentrations in dose dependent manner. However, the strongest antitumor effect was referred to E. aphylla ethanol extract followed by E. foeminea methanol one. As 10% and 28% cell viability at 400 µg/ml was obtained, respectively. The observed acquired antitumor effect could be achieved via anti-proliferative effect (cytostatic) rather than cytotoxic effect.It worth noting that the traditional Palestinian medicinal plants can serve as a source for the discovery of new anticancer agents. However, further research is needed to fully characterize their activity to exclude or include the activity of additional compounds in the plant’s species under study in this research.

In vitro inhibition of sheep liver glutathione-s-transferases activity by different extracts from ephedra aphylla and ephedra foeminea

Glutathione-S-transferases (GSTs) play an important role in the detoxification of chemicals that may lead to mutagenic or cytotoxic effects. In this study, we aimed to find the effect of aqueous, methanol and ethanol extracts of both Ephedra aphylla and Ephedra foeminea on the ac-tivity of the purified sheep liver GST at the following different concentrations (0.1, 0.2, 0.4, 0.6, 0.8 and 1 mg/ml) spectrophotometrically using 1-chloro- 2, 4- dinitrobenzene (CDNB) as substrate. The obtained results revealed that all examined extracts types of Ephedra species under study ma-nifested inhibitory effect on GSTs activity at all screened concentrations. The inhibitory effect on GST activity by the alcoholic extracts of the two species under investigation at all tested concentra-tions was more than the extracts when compared to controls. Nevertheless, this was more pro-nounced in E. aphylla rather than in E. foeminea. It is worth mentioning that all the GSTs inhibito-ry effects displayed dose dependent manner under all studied extracts. On that account, the ob-tained results in this research confirms the medicinal importance of both Ephedra species under study, which provides the possibility of their use in whatever benefit human health in general and combat GST-induced resistance in drug resistant tumors in particular

In vitro Effect of Camel Whey on Copper Inactivated

Glutathione-S-Transferases

The main aim of the present study was to determine the in vitro effect of some heavy metals in general and copper in particular on the activity of sheep liver glutathione-s-transferases (GSTs). Besides that, the in vitro effect of camel whey on copper inactivated GSTs was also studied in this research. The effect of heavy metals zinc, nickel, cobalt, manganese, copper and camel whey on GSTs activity was studied by spectrophotometric method using 1-chloro-2,4-dinitro-benzene (CDNB) as substrate. Results showed that copper ion had the most potent inhibitory effect on GSTs activity when compared to other studied heavy metals as it directly reduced the enzyme residual activity to reach 29.3% at 2 mM concentration. In addition to that, the further increase in incubation time between heavy metals and GSTs lead to an increase in inhibitory behavior of all heavy metals under study. Furthermore, the role of camel whey on copper inactivated GSTs was clearly showed that it can reduced the inhibitory effect of copper on enzyme activity prior to its addition. The acquired residual activity after direct whey addition increased from 0.76% to 16.48% at 2 mM copper concentration. Moreover, the pre-incubation of GSTs with camel whey for a certain period caused better effect than its direct addition. Hence, the residual activity of GSTs increased from approximately zero to 38.5% at 2 mM. In conclusion, camel whey play an important role in the activation of copper inactivated GSTs.

Effect of camel whey immunoglobulins concentrate on

the activity of sheep liver glutathione-s-transferases

The aim of this study was to find out the effect of camel whey immunoglobulins on the activity of glutathione-stransferases (GST). The effect of camel whey immunoglobulins was tested at variable conditions including their concentration effect, their preincubation effect and the effect of pH and temperature on their activity. Moreover, the effect of camel whey

immunoglobulins on the thermal and pH stability of GST was also examined. Furthermore, kinetic parameters of GST were studied in the presence of camel whey immunoglobulins. The observed results showed that camel whey immunoglobulins were not affected by different pH values. Interestingly, milk immunoglobulins preserved the stability and the activity of GST enzyme except in the strong acidic and strong alkaline media. Camel whey immunoglobulins were more stable at 40 ºC and their effect on GST activity declined after 60 ºC. It was clearly noticed that the pre-incubation of GST enzymes with immunoglobulins for a certain period caused better effect than their direct addition. GSH Kinetic results showed that camel whey increased Vmax and Km values. In conclusion, camel whey immunoglobulins play an important role in the activation of GST enzymes even at different conditions.

Cytotoxic potential of camel whey and milk on cervix cancer (HeLa) cell line

Camel milk is an important nutritional source that historically been used in the treatment of cancer. Therefore, the main aim of the present study is to determine the in vitro anticancer effect of both camel milk and whey against cervix cancer (HeLa) cells. To perform that, skimmed milk as well as whey immunoglobulins concentrate samples were prepared at different concentrations (0, 1, 2.5, 5, 7.5 and 10 mg/ml). Then, the in vitro effect of the prepared concentrations on HeLa cells morphology and growth was investigated by tissue culture technique. Moreover, the anticancer activity of camel milk and whey against HeLa cells was estimated by the 3-[4, 5-dimethylthiazole-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) assay. The obtained results displayed that both camel milk and whey reduced the viability of HeLa cells specially at 7.5 and 10 mg/ml. In addition, the viability of treated HeLa cells reduced after addition of both camel milk and whey to approximately 15% in a concentration dependent manner. In conclusion, this study showed the in vitro cytotoxic effect of camel milk and whey as they inhibited the growth of HeLa cells.

Antibacterial activity of camel whey in

combination with various medicinal plant extracts

The aim of the present study was to evaluate the antibacterial potential of ethanol and water extracts from Ballota undulata, Ruta chalepensis and Urtica urens in combination with camel whey proteins. The bioactivity of all samples was tested against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. Disk diffusion method was used to examine the antibacterial potential for all combinations. For further antibacterial investigation, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were measured by micro-broth dilution method. The obtained results

showed that ethanol extracts of B. undulata and R. chalepensis were the most active. Synergistic antibacterial activity was observed for whey and ethanol extracts of B. undulata and R. chalepensis. The MIC and MBC values for these two combinations against S. aureus were equal to 1.56 and 12.5 mg/ml respectively. The obtained results form a platform for further studies on other combinations between camel whey and other plant extracts.

Antimicrobial activity selected copper complexes

Efficacy of different copper complexes against different bacterial strains, including the gram-positive bacterium Staphylococcus aureus and the gram-negative bacterium Escherichia coli. The disk diffusion method was used to determine the susceptibility of different bacterial strains to the examined copper complexes at 0.5 mg/ml. The studied strains produced different responses to different complexes. The most sensitive strains were S. aureus and E. coli. The micro-broth dilution method was used to measure the minimum inhibitory concentration (MIC) and then the minimum bactericidal concentration (MBC) for all copper complexes under study against the two strains. Among the studied complexes, MD showed the best bacteristatic effect as it worked against S. aureus at 62.5 µg/ml. But the best bactericidal agent in this study is ME as it works at 125 µg/ml against S. aureus. Further studies should be done to determine the mechanisms of the antimicrobial activity of copper complexes.

Synthesis, characterization and antimicrobial activity of Copper Oxide nanoparticles

Copper nanoparticles (Cu2O-NPs) were prepared in this work by two methods, including microwave and stirring preparation. The broth micro-dilution method and minimum bactericidal concentration were applied to determine the antibacterial activity of Cu2O-NPs against Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa , Klebsiella pneumoniae and Methicillin resistant Staphylococcus aureus (MRSA). Microwave preparation of Cu2O-NPs exhibited antimicrobial activity against Gram-positive S. aureus (MIC = 125µg/ml). While stirring preparation of Cu2O-NPs produced antimicrobial activity against MRSA and E. Coli (MIC=125 µg/ml) .On the other hand, the stirring preparation of Cu2O-NPs exhibited the best bioactivity against E. coli at the minimum bactericidal concentration (MBC=125 µg/ml).

The effect of some medicinal plants on methicillin resistant Staphylococcus aureus

Based on the importance of herbal plants in medicine as antibacterial agents, this study was conducted to evaluate the antibacterial efficacy of aqueous, ethanol and methanol extracts of three medicinal plants against bacteria. Agar well diffusion method was performed to examine the antibacterial activity of all the studied plant extracts. Micro-broth dilution method was used to measure the minimum inhibitory concentration (MIC) then the minimum bacteriocidal concentration (MBC) for all active extracts. Fractionation and characterization of medicinal plants active components may enhance the pharmaceutical industry of new drugs against bacteria.

Antibacterial Activity of Selected Palestinian Wild Plant Extracts

against Multidrug-Resistant Clinical Isolate of Streptococcus pneumoniae

Streptococcus pneumoniae is one of the most important bacterial pathogens that affect respiratory system. The occurrence of multidrugresistant strains of S. pneumoniae necessitates the discovery of new classes of anti-pneumonia drugs. As some of the medicinal plants and their extracts have antibacterial activity, we aimed to investigate the antibacterial potential of five Palestinian wild medicinal plants including Echinops adenocaulos, Parietaria judaica, Urtica urens, Verbascum fruticulosum and Vitex agnus-castus against multidrug-resistant strain of S. pneumoniae, which was screened for its antibiotic resistance profile showing 67% resistance of the tested antibiotics. In this research micro-broth dilution method was used to study the inhibitory activity of ethanol, distilled water and Zamzam water extracts of all plants under investigation. The obtained results showed that all Zamzam extracts of the five studied plant species inhibited the growth of S. pneumoniae. It was clearly noticed that there was a difference in the antibacterial activity between distilled water and Zamzam water extracts recording higher activity in the Zamzam water extract of some of the studied plant species. Verbascum fruticulosum distilled water extract was the most effective one with MIC equal to 0.195 mg/ml. In general, both water extracts were more bioactive than the ethanol extracts for all the examined plants. These results confirmed the possibility of using these plant species in medicine and pharmaceutical industry of new drugs for the treatment of multidrug-resistant clinical isolates of S. pneumoniae.
